| Synthesis | Ethyl 2-(6-oxo-1,6-dihydropyrimidin-5-yl)acetate (1107) (0.868 g, 4.77 mmol) was mixed with phosphorus oxychloride (POCl3) (6 mL) under nitrogen protection and heated to reflux. The reaction was maintained for 5 min and then cooled to room temperature. The reaction mixture was slowly poured into water (300 mL) and the aqueous phase was extracted with dichloromethane (DCM) (3 x 100 mL). The organic phases were combined, washed with saturated saline (100 mL), dried over anhydrous magnesium sulfate (MgSO4), filtered and concentrated under reduced pressure to afford the target compound 4-chloro-5-pyrimidine ethyl acetate (1108) (0.885 g, 93% yield) as a light yellow oil.1H NMR (400 MHz, CDCl3) δ 8.93 (s, 1H), 8.61 (s , 1H), 4.21 (q, J = 7.1 Hz, 2H), 3.76 (s, 2H), 1.28 (t, J = 7.1 Hz, 3H).LCMS method C: retention time 5.09 min; m/z 201.0, 203.1 [M+H]+. | | References | [1] Patent: WO2012/110773, 2012, A1.
