(2) Via a novel aromatization process, this bicyclic frame can be converted into a series of 5-alkoxytryptamine derivatives including serotonin, melatonin and bufotenin. | | Synthesis | In a dry Schlenk tube, [Rh(cod)Cl]2 (9.9 mg, 20.0 mol), (R)-Binap (27.4 mg, 44.0 mol), and Durum (20.0 mg, internal standard) were dissolved in CH2Cl2 (8.0 mL); then, 2 mmol of the substrate was added to the solution at room temperature under nitrogen. After stirring for 1 min, CF3CO2Ag (8.8 mg, 40.0 mol) was added to the mixture. The solution was stirred at 25C under nitrogen and monitored by GC or HPLC to determine the conversion and ee values. After evaporation of the solvent, the residue was purified by chromatography using EtOAc/hexane (20/80) as eluent to give the product 1,4-cyclohexanedione monoethylene glycol ketal. | | Purification Methods | Recrystallise it from pet ether.
