TAE electrophoresis buffers are the most commonly used buffers for DNA agar-gel electrophoresis and are also used for
non-denatured RNA agar-gel electrophoresis. Double-stranded DNA tends to move faster in TAE than in other buffers, but can also be  immobile during long electrophoresis periods due to buffer ion depletion. Buffer circulation or buffer replacement during
prolonged electrophoresis can compensate for the lower Chemicalbook buffering capacity. Dilute concentrated TAE buffer to produce  1mMTAE buffer containing 40mMTris acetate and 1mMEDTA at a pH of 8.3. The 1mMTAE buffer can be used either in agarose gels or as  an electrophoresis buffer. For maximum resolution, the applied voltage is recommended to be less than 5V/cm (unit distance between electrodes).
