Cytoglobin (CYGB), a member of the globin family, was discovered in 2002 as a ubiquitously expressed protein with potential roles in oxygen metabolism, redox homeostasis, and nitric oxide regulation. Structurally, it shares a conserved globin fold but features a unique hexacoordinate heme-binding structure, distinguishing it from hemoglobin and myoglobin. CYGB is predominantly expressed in fibroblasts, smooth muscle cells, and certain tissue-specific stromal cells, though its exact physiological function remains debated. Proposed roles include oxygen storage/delivery, scavenging reactive oxygen species (ROS), and modulating collagen synthesis, particularly under hypoxic or fibrotic conditions.

CYGB antibodies are critical tools for investigating its involvement in pathological processes. Studies link CYGB dysregulation to cancer progression, fibrosis (e.g., liver, lung), and cardiovascular diseases, with both tumor-suppressive and pro-survival functions reported depending on context. Antibodies targeting CYGB enable detection via Western blot, immunohistochemistry (IHC), and immunofluorescence (IF), helping map its tissue distribution and subcellular localization (nuclear vs. cytoplasmic). Commercial antibodies vary in specificity, necessitating validation using knockout controls. Recent research explores CYGB's interaction with signaling pathways (e.g., HIF-1α, TGF-β) and its potential as a biomarker or therapeutic target. However, inconsistent findings across studies highlight the need for standardized antibody protocols to clarify CYGB's multifaceted roles in health and disease.
