The dopamine receptor D5 (DRD5) is a member of the dopamine receptor family, classified as a G protein-coupled receptor (GPCR) within the D1-like subfamily (D1 and D5). It is encoded by the DRD5 gene and is distinguished by its high basal activity and ligand-independent signaling. Structurally, DRD5 shares homology with DRD1 but has a longer third intracellular loop, which may influence G protein coupling. Functionally, DRD5 primarily couples to Gαs proteins, activating adenylate cyclase and increasing intracellular cAMP levels upon dopamine binding. It is expressed in key brain regions, including the hippocampus, prefrontal cortex, and hypothalamus, implicating roles in cognition, emotion, and neuroendocrine regulation.

DRD5 antibodies are essential tools for studying the receptor's localization, expression levels, and signaling mechanisms. They are widely used in techniques like Western blotting, immunohistochemistry (IHC), and flow cytometry to visualize DRD5 in tissues or cultured cells. Specificity validation via knockout controls or blocking peptides is critical due to potential cross-reactivity with DRD1. Research involving DRD5 antibodies has explored its involvement in neurological and psychiatric disorders, such as schizophrenia, attention-deficit/hyperactivity disorder (ADHD), and addiction, as well as its interplay with other dopamine receptors. Recent studies also investigate DRD5's role in non-neural tissues, such as renal and immune systems, broadening its physiological relevance.
