CD8A antibodies target the CD8α chain, a critical surface glycoprotein expressed primarily on cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. The CD8 molecule functions as a co-receptor for T-cell receptors (TCRs), binding to major histocompatibility complex class I (MHC-I) molecules on antigen-presenting cells to stabilize TCR-peptide-MHC interactions and enhance T-cell activation. Structurally, CD8 exists as a heterodimer (CD8αβ) or homodimer (CD8αα), with the α-chain encoded by the *CD8A* gene.

CD8A antibodies are widely used in research and diagnostics to identify and isolate CD8+ T cells, which play central roles in antiviral and antitumor immunity. In flow cytometry, these antibodies help characterize T-cell subsets, monitor immune responses, and assess pathologies like autoimmune diseases or immunodeficiencies. They also enable spatial analysis of CD8+ T-cell infiltration in tumor tissues via immunohistochemistry, a prognostic marker in cancer immunotherapy. Therapeutic applications include bispecific antibodies engaging CD8 for targeted cytotoxicity and CAR-T cell engineering. Commercially available clones (e.g., OKT8. SK1) are validated for specificity across human and model species, with formats optimized for diverse sample types (fresh, frozen, or FFPE tissues). Recent advancements in single-cell technologies and multiplex assays have further expanded their utility in deciphering immune microenvironments and predicting responses to checkpoint inhibitors.
