NVG-291 is a synthetic peptide analogue of intracellular sigma peptide (ISP), used to regulate the proteoglycan receptor PTP sigma. It is a peptide composed of 35 amino acids. NVG-291 can exert its effect by inhibiting the interaction between chondroitin sulfate proteoglycans (CSPGs) and PTP σ, both of which inhibit the differentiation of oligodendrocyte precursor cells (OPCs) and myelin regeneration. In studies utilizing lysophosphatidylcholine (LPC) and experimental autoimmune encephalomyelitis (EAE) models, the application of ISP is associated with increased survival, proliferation, and maturation of OPCs, which helps improve axonal myelin regeneration.
