Fasinumab (development code REGN475) is an investigational, fully human monoclonal antibody developed for the management of chronic pain, specifically osteoarthritis (OA) pain and chronic low back pain. It functions as a highly selective and potent nerve growth factor (NGF) inhibitor. By binding to and neutralizing NGF in the bloodstream and tissues, fasinumab prevents NGF from interacting with its receptors, tropomyosin receptor kinase A (TrkA) and the p75 neurotrophin receptor (p75NTR), on sensory neurons. This blockade inhibits the NGF-mediated signaling cascade that normally sensitizes pain pathways, leading to a reduction in pain perception.
