Sacituzumab is an antibody-drug conjugate (ADC) targeting Trop-2 (trophoblast cell surface antigen 2), a transmembrane glycoprotein overexpressed in many epithelial cancers. It is linked to the cytotoxic payload SN-38 (a topoisomerase I inhibitor). Upon binding to Trop-2, the ADC is internalized, releasing SN-38 to induce DNA damage and tumor cell death. It is approved for the treatment of metastatic triple-negative breast cancer and urothelial carcinoma.
