A1.2 Step 2:
To a solution of 1-benzyl-1H-pyrazole (5.1 g, 32.3 mmol) in acetic anhydride (11.0 mL, 116 mmol) was added sulfuric acid (0.17 mL, 3.23 mmol). The solution was refluxed for 4 hours. The reaction was cooled to r.t., and water was added. The mixture was cooled to 0°C and basified with NaOH to pH >10. The product was extracted with DCM, and the combined organic layers were washed with brine, dried over Na2SO4, filtered, and concentrated in vacuo. The crude material was purified by silica gel chromatography eluting with 10-40% EtOAc in hexanes to provide 1-(1-benzyl-1H-pyrazol-4-yl)ethan-1-one-1 (4.21 g, 21.0 mmol, 64% yield) as a beige solid.1H NMR (400 MHz, Chloroform-d) δ ppm 7.93 (s, 1H), 7.84 (s, 1H), 7.46 - 7.31 (m, 3H), 7.29 - 7.24 (m, 2H), 5.31 (s, 2H), 2.41 (s, 3H).LC/MS (ESI+) m/z = 201.1 [M+H]+.
