A.2 Step 2: (5)-tert-butyl 3-(cyanomethyl)pyrrolidine-1-carboxylate
100 198j To a solution of (5)-tert-butyl 3-(tosyloxymethyl)pyrrolidine- 1 -carboxylate (3.0 g, 8.44 mmol) in DMSO (40 mL) was added NaCN (596 mg, 10.13 mmol) at 100°C overnight. TLC showed the reaction was completed. The mixture was poured into the ice water andextracted with ethyl acetate (100 mL x 2). The organic layers were dried over Na2SO4, filtered and concentrated. The residue was purified by silica gel column chromatography (petroleum ether : ethyl acetate = 100 : ito 4: 1) to give (R)-tert-butyl 3-(cyanomethyl)pyrrolidine-i- carboxylate (1.6 g, 90.16% yield) as a white solid. LC-MS: m/z 155 = [M+H-56j.
