Synthesis of (S)-6-bromo-1-(sec-butyl)-N-((4,6-dimethyl-2-oxo-1,2-dihydropyridin-3-yl)methyl)-3-methyl-1H-indole-4-carboxamide from 2-(piperazin-1-yl)pyridine-5-boronic acid pinacol ester and (S)-6-bromo-1-(sec-butyl)-N-((4,6-dimethyl-2-oxo-1,2- dihydropyridin-3-yl)methyl)-3-methyl-6-(6-(piperazin-1-yl)pyridin-3-yl)-1H-indole-4-carboxamide was prepared in the following general procedure: in a 100 mL three-necked flask (S)-6-bromo-1-(sec-butyl)-N-((4,6-dimethyl-2-oxo-1,2-dihydropyridin-3-yl)methyl)-3-methyl-1H-indole-4-carboxamide was added. indole-4-carboxamide (365 mg, 0.82 mmol), pinacol ester of 2-(piperazin-1-yl)pyridine-5-boronic acid (309 mg, 1.07 mmol, 1.3 eq.), potassium phosphate (522 mg, 2.46 mmol, 3 eq.), water, and 1,4-dioxane solvent. Subsequently, [1,1'-bis(diphenylphosphino)ferrocene]palladium(II) dichloride dichloromethane complex (53.9 mg, 0.066 mmol) was added under nitrogen protection and the reaction mixture was heated to 90 °C for the reaction. Upon completion of the reaction, the target product was purified to give 400 mg in 92% yield.
