4.1.2.2. General procedure for synthesis of 2-substituted imidazole,method B.
General procedure: A suspension of NH4HCO3 (13.8 g, 174.6 mmol) and 40%glyoxal solution (10.9 mL,87.3 mmol) in water (9 mL) was treateddropwise with appropriate aldehyde (9.4 mL, 87.3 mmol) at 0 C.The mixture was stirred at 25 C for 24 h. The mixture wasconcentrated under reduced pressure. The residuewas diluted withCH2Cl2 and washed with brine. The organic layer was dried overanhydrous MgSO4, filtered and concentrated. The product waspurified by column chromatography (CH2Cl2/MeOH 25:1) toobtain the title compound.The following compound was synthesized using method B: 2-isobutyl-1H-imidazole (12c), 2-cyclopropyl-1H-imidazole (12d),2-cyclopentyl-1H-imidazole (12e), 2-cyclohexyl-1H-imidazole(12f).
