Steps:
Synthesis of 2, 4-dichloro-5-methyl-5H-pyrrolo[3,2-d]pyrimidine (IIa).
General procedure: A mixture of 2,4-dichloro-5H-pyrrolo[3,2-d]pyrimidine (I, 1.20 g, 6.38 mmol)with iodomethane (1.09 g, 7.66 mmol) and cesiumcarbonate (Cs2CO3, 1.04 g, 3.20 mmol) in N,Ndimethylformamide(DMF, 10 mL) was stirred atroom temperature for 24 h (TLC). Then, the mixturewas added ethyl acetate (EtOAc, 40 mL) and washedfive times with saturated saline, the EtOAc was driedover anhydrous sodium sulfate (Na2SO4) and filtered.After removing the solvent under vacuum, the residuewas purified by silica gel chromatography to get compound(IIa) as a yellow solid.
