2 3-(4-Aminophenyl)piperidine-2,6-dione
To a solution of 3-(4-nitrophenyl)piperidine-2,6-dione (55 g, 235 mmol) in ethanol (550 mL, 0.43 M) was added a solution of ammonium chloride (75.4 g, 1.41 mol, 6 equiv.) in water (220 mL) and iron powder (65.6 g, 1.17 mol, 5 equiv.). The mixture was stirred at 80 °C. After 1 h the mixture was cooled to 60 °C and filtered. The solids were washed with acetonitrile (1L) at 60°C and filtered.The combined filtrates were concentrated to give 3-(4-aminophenyl)piperidine-2,6-dione (33.9 g, 166 mmol, 71% yield) as a light-yellow solid.1H NMR (400 MHz, DMSO-d6) δ 10.73 (s, 1H), 6.85 (d, J = 8.4 Hz, 2H), 6.52 (d, J = 8.4 Hz, 2H), 4.98 (s, 2H), 3.64-3.60 (m, 1H), 2.65-2.56 (m, 1H), 2.47-2.41 (m, 1H), 2.12-1.95 (m, 2H).
