Synthesis of 2-methyl-2-(4-(trifluoromethyl)phenyl)propionic acid
A solution of 2-methyl-2-(4-(trifluoromethyl)phenyl)propionitrile (400 mg, 1.88 mmol) and NaOH (10 M, 1 mL) in EtOH (5 mL) was refluxed at 100°C overnight.
After the mixture was cooled to room temperature, EtOAc (15 mL) was added.
The two layers were separated, and the organic solution was extracted with water (10 mL*3).
The combined aqueous layer was acidified with concentrated hydrochloric acid to pH=2-3. HCl solution.
Then the aqueous layer was extracted with EtOAc (20 mL*3).
The combined organic layer was washed with brine (20 mL*2), dried over anhydrous Na2SO4 and concentrated in vacuo to obtain the desired compound (390 mg, yield 81%) as a white solid. 1HNMR (400 MHz, DMSO-d6): δ 12.56 (s, 1H), 7.70 (d, J= 8.0 Hz, 2H), 7.58 (d, J= 8.0 Hz, 2H), 1.51 (s, 6H).
