9.9-Dimethyl-2-bromoindole was sequentially introduced into a dry 5L four-necked flask.273.2 g (1.0 mol, 99%, 1.0 eq.),O-chloroaniline153.1 g (1.2 mol, 99%, 1.2 eq.),Pd2dba3 catalyst4.58g (0.005mol, 97%, 0.5% eq.),t-Bu3PHBF4 ligand 2.9 g (0.01 mol, 99%, 1% eq.),Potassium tert-butoxide 224.4g (2mol, 98%, 2.0eq.) and xylene 2.8kg,Nitrogen was replaced three times and heated under reflux for 18 h under nitrogen.The reaction of the reaction, 9,9-dimethyl-2-bromoindole, was completely converted by TLC.After the reaction was completed, the temperature was lowered to room temperature, and quenched by adding 500 g of ice water, stirred for 1 hour, and layered.The organic phase is retained; the organic phase is washed twice with 2*250g water;The organic phase is concentrated to recover the solvent xylene; a brown red viscous liquid is obtained.Recrystallization from dichloromethane and n-hexane afforded 219.9 g of pale yellow solid.The yield was 77.6%.
