Reaction Conditions:

Stage #1: 1-chloro-4-pentynewith n-butyllithium in tetrahydrofuran at -78; for 1 h;
Stage #2: with 2-Isopropoxy-4,4,5,5-tetramethyl-1,3,2-dioxaborolane in tetrahydrofuran; for 2 h;
Stage #3: with oxone in acetone at 50; for 12 h;

At -78°C, n-butyllithium (2.4 mL, 2.5 mol / L, 1.2 eq.) Was added dropwise to a solution of 5-chloropentyne (512.8 mg, 1.0 eq.) In tetrahydrofuran (15 mL) for 1 h. Isopropanol pinacol borate (1.2 mL, 1.2 equivalents) was added dropwise to the mixed solution, and the reaction was continued for 2 hours. A 1,4-dioxane solution of hydrogen chloride (1.9 mL, 4 M in dioxane, 1.5 equivalents) was added. After quenching, the reaction was warmed to room temperature. After the solvent was rotary evaporated, it was prepared into an acetone solution (0.3M), and then potassium peroxymonosulfonate solution (0.5M, 4.6107g, 1.5 equivalents) was added, and transferred to 50°C and stirred for 12h. After separation and purification, 676.1 mg of 5-chlorovaleric acid with a purity of 99% was obtained.
