3.B Step B: Preparation of Compound 2-3:
To a solution of tert-butyl 4-(1-methoxy-1-oxopropan-2- yl)piperazine-1-carboxylate (540 mg, 2 mmol) in THF (10 mL) was added LiAlH4 (1.0 mL, 2.5 mol in THF) at 0 °C dropwise. The reaction mixture was stirred at the same temperature for 2 hours. TLC showed that the reaction was complete. The reaction was quenched with EtOAc. The reaction was partitioned between EtOAc and H2O, and the organic layer was washed with brine and dried over Na2SO4. Solvent was removed under vacuum and the residue was purified by silica gel column chromatography (CH2Cl2/MeOH = 20:1) to give tert-butyl 4-(1-hydroxypropan-2-yl)piperazine-1-carboxylate (2-3) as a brown oil (300 mg, yield: 65%).
