Steps:

Serine, threonine, and phenylserine synthesis

The initial velocity of serine, threonine, or phenylserine synthesiswas measured in a 500 μL reaction with the followingbuffer solution components: 5.54 U SHMT, 50 mM phosphatebuffer pH 7.5, 25 mM sodium sulfate, 50 mM glycine,2 mM THF (for serine synthesis only), 50 mM formaldehyde(for serine synthesis), 10 mM acetaldehyde (for threoninesynthesis), 1 M benzaldehyde (for phenylserine synthesis),100 mM β-mercaptoethanol, and 0.3 μM PLP. The mixturewas incubated at various temperatures (50-100 °C) for 5 minbefore the enzymatic reaction was terminated with the additionof 1% TCA.
