The general procedure for the synthesis of 4-chloro-5-iodo-2-methyl-7H-pyrrolo[2,3-d]pyrimidines from 4-chloro-2-methyl-1H-pyrrolo[2,3-d]pyrimidines was as follows: to a solution of 4-chloro-2-methyl-7H-pyrrolo[2,3-d]pyrimidines (400 mg, 2.4 mmol) in methylene chloride (10 mL) was added N-iodosuccinic imide (537 mg, 2.39 mmol). The reaction mixture was stirred at room temperature for 2 hours. Upon completion of the reaction, the organic layer was washed with saturated aqueous sodium sulfite to remove unreacted iodine. The organic layer was dried over anhydrous sodium sulfate, the desiccant was removed by filtration and the filtrate was concentrated under reduced pressure to give a brown solid product. Yield: 330 mg, 1.12 mmol, 47% yield. The product was analyzed by LCMS, m/z 293.8 [M + H]+. | [References]
