General procedure for the synthesis of 6-bromooxazolo[4,5-b]pyridine from 3-hydroxy-2-amino-5-bromopyridine and triethyl orthoformate: 2-amino-5-bromo-3-hydroxypyridine (200 mg, 1.06 mmol) was dissolved in 3 mL of triethyl orthoformate and a catalytic amount of p-toluenesulfonic acid was added. The reaction mixture was heated to reflux for 5 hours. After completion of the reaction, it was cooled to room temperature, diluted with dichloromethane (DCM) and washed with saturated sodium bicarbonate solution. The organic phase was dried with anhydrous sodium sulfate and subsequently concentrated to dryness under reduced pressure. The residue was purified by fast column chromatography to give 212 mg (60% yield) of 6-bromoxazolo[4,5-b]pyridine. | [References]
