Ethyl 4-amino-3-iodobenzoate was synthesized under heating conditions using ethyl p-aminobenzoate and potassium iodide as raw materials and sodium hypochlorite as an oxidant. A series of factors affecting the synthesis of ethyl 4-amino-3-iodobenzoate were investigated. The results showed that at a temperature of 30℃, with 0.01 mol of ethyl p-hydroxybenzoate, 0.01 mol of potassium iodide, 0.02 mol of oxidant, a reaction time of 2 h, and using 5 mL of water and 15 mL of anhydrous ethanol as solvents, the highest yield of ethyl 4-amino-3-iodobenzoate could reach 90.2%. This method is environmentally friendly and pollution-free. This method is green and environmentally friendly. Ethyl 4-amino-3-iodobenzoate, aqueous lithium hydroxide solution, and methanol were added sequentially to a reaction flask, and the mixture was reacted at 30°C for 6 h with stirring. The solvent was removed under vacuum, the pH was adjusted to 3-5 with dilute hydrochloric acid, and the mixture was extracted with dichloromethane (2 × 50 mL). The extracts were combined, concentrated, and purified by silica gel column chromatography [eluent: V(dichloromethane):V(methanol) = 15:1] to obtain 4-amino-3-iodobenzoic acid. |
