The general procedure for the synthesis of 4-amino-2,5,6-trichloropyrimidine from 4-amino-2,6-dichloropyrimidine was as follows: 1 g (6.1 mmol) of 4-amino-2,6-dichloropyrimidine was dissolved in 5 ml of N,N-dimethylformamide (DMF). To this solution 0.98 g (7.32 mmol) of N-chlorosuccinimide (NCS) was added. The reaction mixture was poured into 100 ml of ice water. The precipitate formed was collected by filtration, washed several times with distilled water and dried under vacuum to give 1.4 g (80% yield) of the target product as a white solid. The structure of the product was confirmed by 1H nuclear magnetic resonance (NMR) spectrum (300 MHz, DMSO-d6): δ= 8.24 (d, 2H). | [References]
