GENERAL STEPS: Vilsmeier's reagent was prepared by slowly adding 0.262 mL (2.810 mmol) of phosphorus oxychloride (POCl3) dropwise to 0.262 mL (3.383 mmol) of dry N,N-dimethylformamide (DMF) dissolved in 0.4 mL of chloroform (CHCl3) under stirring at 0-5 °C. Subsequently, a solution of 6-(trifluoromethyl)imidazo[2,1-b]thiazole (100 mg, 0.520 mmol) dissolved in 3 mL of chloroform (CHCl3) was slowly added dropwise to the above mentioned Vilsmeier reagent under continuous stirring and cooling conditions. The reaction mixture was first stirred at room temperature for 3 hours and then warmed up to reflux state and kept for 39 hours. After completion of the reaction, it was cooled to room temperature and the reaction mixture was poured into ice water and extracted with dichloromethane (DCM, 3×). The organic phases were combined, dried with anhydrous magnesium sulfate (MgSO4), filtered and concentrated under reduced pressure to give the crude product. Purification by fast column chromatography (n-heptane/ethyl acetate, 8:1) afforded 6-(trifluoromethyl)imidazo[2,1-b]thiazole-5-carboxaldehyde as an oil (360 mg, 52% yield).1H-NMR (CDCl3) data: δ= 7.30 (d, 2H); 8.45 (s, 1H); 10.05 (s, 1H).
