b) Synthesis of ethyl 12-bromododecanoate
Ethyl 12-hydroxydodecanoate (1.65 g, 6.7 mmol) was dissolved in dichloromethane (20 mL) under nitrogen protection in a 100 mL two-necked round-bottomed flask equipped with a reflux condenser tube and a magnetic stirrer. Triphenylphosphine (PPh3, 1.93 g, 7.4 mmol) and N-bromosuccinimide (NBS, 1.6 g, 7.0 mmol) were added sequentially. The reaction mixture was stirred under reflux conditions for 24 hours. Upon completion of the reaction, the reaction solution was concentrated using a rotary evaporator and the resulting crude product was purified by silica gel column chromatography with petroleum ether/ethyl acetate (5:1, v/v) as eluent. The final isolation afforded 1.92 g of ethyl 12-bromododecanoate in 92% yield as a light yellow oil. | [References]
