向5-碘尿苷(5.0 g,14.1 mmol)的无水DMF(40 mL)溶液中,在氮气保护下加入CuI(0.20 g,1.05 mmol)和Pd(PPh3)4(0.41 g,0.35 mmol)。搅拌10分钟后,依次加入三乙胺(4.0 mL,28.6 mmol)和2,2,2-三氟-N-(丙-2-炔-1-基)乙酰胺(5.4 g,35.7 mmol),将反应混合物在室温下搅拌过夜。反应完成后,在真空下除去所有挥发物,残余物通过硅胶快速色谱法纯化[洗脱剂:EtOAc/MeOH(0-15%)],得到目标产物2,2,2-三氟-N-(3-(1-((2R,4S,5R)-4-羟基-5-(羟甲基)四氢呋喃-2-基)-2,4-二氧基-1,2,3,4-四氢嘧啶-5-基)丙-2-炔基)乙酰胺,为黄色固体(4.1 g,收率76%)。1H-NMR(DMSO-d6)δ: 11.68(br s, 1H, NH), 10.02(s, 1H, NH), 7.94(s, 1H, H-6), 6.11(t, J = 7.2 Hz, H-1'), 5.29(d, J = 0.4 Hz, 1H, OH), 4.21(m, 3H, H-3', NCH2), 3.71-3.83(m, 2H, CH2-5'), 2.15-2.19(m, 1H, H-2'), 2.03-2.08(m, 1H, H-2')。质谱(ESI): 计算值C14H14F3N3O6(M + Na)+ 400,实测值400。
