将2'-脱氧-2'-氟尿苷(3.00 g,12.2 mmol)溶解于无水吡啶中,重复进行减压浓缩操作3次以实现脱水共沸蒸馏。随后,在氩气保护下,将反应混合物溶解于无水吡啶(120 mL)中,加入4,4'-二甲氧基三苯甲基氯(4.55 g,13.4 mmol),室温搅拌反应3小时。反应完成后,向反应混合物中加入乙酸乙酯(150 mL)和水(60 mL),进行分层。有机层依次用5%碳酸氢钠水溶液(20 mL×3)、水(20 mL)和饱和食盐水(20 mL)洗涤,随后用无水硫酸钠干燥。过滤后,蒸发去除溶剂,残余物通过硅胶柱色谱纯化(洗脱剂:己烷/乙酸乙酯=50:50至0:100(v/v),含1%三乙胺)。收集含目标产物的馏分,浓缩后得到1-((2R,3R,4R,5R)-5-((双(4-甲氧基苯基)(苯基)甲氧基)甲基)-3-氟-4-羟基四氢呋喃-2-基)嘧啶-2,4(1H,3H)-二酮(8.48 g,定量收率)。
