以5-甲氧基烟腈为原料合成3-氰基-5-羟基吡啶的一般步骤:将1g 5-甲氧基烟腈(7.46mmol)与8.62g盐酸吡啶混合,在200℃下加热反应2小时。反应完成后,将粗产物用水稀释,并用乙醚多次萃取。水相通过加入碳酸氢钠进行碱化后,再次用乙醚萃取。合并有机相,经无水硫酸钠干燥后,减压浓缩,得到850mg米色固体5-羟基烟腈,收率95%。产物经LC-MS分析:m/z 120.94(M+H+)。1H NMR(400MHz, DMSO-d6):δ 10.79(s, 1H, OH),8.46(s, 1H, Harom.),8.42(s, 1H, Harom.),7.60(s, 1H, Harom.)。
