在250 mL三颈烧瓶中,将2,4-二氯嘧啶(5.0 g,33.56 mmol)溶于1,4-二恶烷和水的混合溶剂(4:1,50 mL)中,加入吡啶-3-硼酸(4.95 g,40.27 mmol)、碳酸钾(9.28 g,67.12 mmol)和Pd(dppf)Cl2(2.45 g,3.36 mmol)。用氩气置换体系三次,逐渐加热至90℃,反应4小时。反应完成后,在减压下浓缩体系以除去大部分溶剂,加入乙酸乙酯(150 mL)和水(100 mL)进行萃取分离,再用乙酸乙酯(80 mL)萃取水相。合并有机相,依次用水(80 mL×2)和饱和食盐水(80 mL×2)洗涤,有机相用无水硫酸钠干燥3小时,过滤并浓缩得到粗产物。粗产物用石油醚和乙酸乙酯(6 mL)混合溶剂搅拌1小时,过滤,真空干燥,得到4.8 g 2-氯-4-(3-吡啶基)嘧啶,收率为74.6%,HPLC纯度大于95%。
