在500 mL三口烧瓶中，加入300 mL无水乙醇，依次加入80 g (0.611 mol) N-乙酰-β-丙氨酸和86.1 g (0.555 mol) L-组氨酸，随后加入98 g (0.582 mol) N,N'-二甲基碳二亚胺(DMC)。将反应混合物加热至40°C，保持搅拌反应3小时。反应完成后，将反应液冷却至5°C，析出固体，抽滤收集固体产物。用150 mL无水乙醇洗涤滤饼，真空干燥后得到145 g (S)-2-(3-乙酰氨基丙酰胺)-3-(1H-咪唑-4-基)丙酸(IV)，收率97.9%，HPLC纯度>98.8%。
