以6-乙酰氧基-4-氯-7-甲氧基喹唑啉为原料合成4-氯-7-甲氧基-6-醇的一般步骤:将4-氯-7-甲氧基喹唑啉-6-基乙酸酯(按照WO01/66099实施例25-5所述方法制备,10.1 g,40 mmol)悬浮于6 N甲醇氨溶液(200 mL)中,室温搅拌90分钟。反应完成后,于真空条件下蒸发除去溶剂。向残余物中加入水,过滤所得悬浮液。收集的固体依次用水和乙醚洗涤,随后在高温下干燥。最后,在五氧化二磷存在下进行真空干燥,得到4-氯-7-甲氧基喹唑啉-6-醇(7.9 g,收率94%)。核磁共振氢谱(DMSO-d6)数据如下:δ 4.02(s,3H),7.40(s,1H),7.43(s,1H),8.81(s,1H)。
