向250 mL圆底烧瓶中加入50.0 g (0.42 mol)对甲基苯甲醛、47.6 g (0.46 mol)丙二酸和64.2 g (0.83 mol)乙酸铵,溶于乙醇中。在搅拌下,将反应混合物加热至80-90°C回流反应7.5小时。反应完成后,将混合物冷却至0-5°C并搅拌30分钟,随后过滤,得到51.4 g 3-氨基-3-(对甲苯基)丙酸(外消旋混合物),为白色粉末(基于对甲基苯甲醛的分离产率为68.9%)。 3-氨基-3-(对甲苯基)丙酸(外消旋混合物)的物理性质如下: 1H-NMR (δ, ppm, D2O + DCl): 2.30 (s, 3H), 3.04 (dd, 1H, J = 17.1, 6.8 Hz), 3.20 (dd, 1H, J = 17.1, 7.3 Hz), 4.74 (dd, 1H, J = 7.3, 6.8 Hz), 7.29 (d, 2H, J = 8.3 Hz), 7.36 (d, 2H, J = 8.3 Hz). 13C-NMR (δ, ppm, D2O + DCl): 23.4, 40.7, 54.4, 130.0, 133.0, 135.0, 143.1, 176.3. MS (EI) m/z: 179 (M+). MS (CI, i-C4H10) m/z: 180 (MH+). 元素分析: 计算值 C, 67.02%; H, 7.31%; N, 7.82%. 实测值 C, 67.05%; H, 7.40%; N, 7.66%.
