以5-溴-7-氮杂吲哚为原料合成5-溴-3-碘-7-氮杂吲哚的一般步骤:制备5-溴-3-碘-1H-吡咯并[2,3-b]吡啶(中间体A)[0442]。在500 mL丙酮中搅拌溶解5-溴-1H-吡咯并[2,3-b]吡啶(10 g,50.76 mmol),随后加入N-碘代琥珀酰亚胺。将反应混合物于室温下搅拌20分钟。反应完成后,将产物粉碎,过滤收集白色固体,并用100 mL丙酮洗涤。所得固体经真空干燥,得到5-溴-3-碘-1H-吡咯并[2,3-b]吡啶(16.34 g,100%收率),呈浅黄色粉末。1H NMR (DMSO-d6, 300 MHz) δ 8.51 (d, J = 2.1 Hz, 1H), 8.22 (s, 1H), 8.02 (d, J = 1.2 Hz, 1H), 8.00 (d, J = 5.1 Hz, 2H), 7.44 (dd, J = 8.7 Hz, 0.6 Hz, 2H), 2.35 (s, 3H); MS ESI (m/z): 322/324 [M + H]+, 计算值322。
