3-(2-氯-6-氟苯基)-5-甲基异恶唑-4-羧酸是一种抗生素氟氯西林钠的关键中间体,也称为氟氯西林-杂质D,可用于实验室研发和有机合成过程中。
制备3-(2-氯-6-氟苯基)-5-甲基异恶唑-4-异恶唑甲酰氯:
3-(2-氯-6-氟苯基)-5-甲基异恶唑-4-羧酸∶双(三氯甲基)碳酸酯∶四甲基脲=1∶0.33∶0.02(摩尔比),3-(2′-氯-6′-氟苯基)-5-甲基-4-异恶唑甲酸投料量为25.55g(100mmol),双(三氯甲基)碳酸酯投料量为9.80g(33mmol),催化剂用量为0.232g(2mmol),甲苯用量为3-(2′-氯-6′-氟苯基)-5-甲基-4-异恶唑甲酸质量的10倍。
3-(2′-氯-6′-氟苯基)-5-甲基-4-异恶唑甲酸、四甲基脲和甲苯加到反应釜中,搅拌均匀后,在室温下45分钟内滴加双(三氯甲基)碳酸酯的甲苯溶液,同时开启氯化氢吸收系统,然后升温到110℃,回流反应2小时,反应结束后减压蒸馏回收甲苯,最后在0.667KPa下收集168~170℃的馏分,冷冻下固化。
