生产方法
以3-乙基哌啶-4-酮和二碳酸二叔丁酯为原料合成3-乙基-4-氧代哌啶-1-甲酸叔丁酯的一般步骤如下:将3-乙基哌啶-4-酮(2.90g,10.2mmol)悬浮于6N盐酸(85ml)中,回流反应16小时。反应完成后,浓缩反应液,并通过与异丙醇共沸三次去除残余水分,得到黄色固体。将该固体溶解于1:1的二氯甲烷与甲醇混合溶剂(50ml)中,依次加入三乙胺(3.08g,4.3ml,30.5mmol)和二碳酸二叔丁酯(3.32g,15.2mmol)。将反应混合物在23℃下搅拌16小时,随后浓缩反应液。向浓缩物中加入0.5N氢氧化钠溶液(50ml),并用二氯甲烷萃取。合并有机相,用无水硫酸镁干燥,过滤后浓缩。最后,通过硅胶柱色谱法(洗脱剂:10%乙酸乙酯-己烷)纯化,得到1.92g(8.46mmol,收率83%)目标产物3-乙基-4-氧代哌啶-1-甲酸叔丁酯,为无色油状物。质谱(ESI+):m/z 228 [M+1]+。
