(３) 本品を乾燥し，赤外吸収スペクトル測定法〈2.25〉の臭化カリウム錠剤法により試験を行い，本品のスペクトルと
本品の参照スペクトルを比較するとき，両者のスペクトルは
同一波数のところに同様の強度の吸収を認める． | | 定量法 | 本品を乾燥し，その約0.35gを精密に量り，無水酢酸
／酢酸(100)混液(7：3)80mLに溶かし，0.1mol/L過塩素酸で
滴定〈2.50〉する(電位差滴定法)．同様の方法で空試験を行い，
補正する． | | 純度試験 | (１) 塩化物〈1.03〉 本品0.6gをるつぼにとり，炭酸ナト
リウム試液2.5mLに溶かし，水浴上で加熱して蒸発乾固した

希硝酸6mL及び水を加えて50mLとする(0.015％以下)．
(２) 重金属〈1.07〉 本品1.0gをとり，第2法により操作し，
試験を行う．比較液には鉛標準液2.0mLを加える(20ppm以
下)．
(３) 類縁物質 本品0.10gをアンモニア試液のメタノール
溶液(1→100)10mLに溶かし，試料溶液とする．この液1mL
を正確に量り，アンモニア試液のメタノール溶液(1→100)を
加えて正確に25mLとする．更にこの液1mLを正確に量り，
アンモニア試液のメタノール溶液(1→100)を加えて正確に
20mLとし，標準溶液とする．これらの液につき，薄層クロ
マトグラフィー〈2.03〉により試験を行う．試料溶液及び標
準溶液10μLずつを薄層クロマトグラフィー用シリカゲルを
用いて調製した薄層板にスポットする．次にアセトニトリル
／水／アンモニア水(28)混液(90：10：1)を展開溶媒として
約15cm展開した後，薄層板を風乾する．これに噴霧用ドラ
ーゲンドルフ試液を均等に噴霧し，更に過酸化水素試液を均
等に噴霧するとき，試料溶液から得た主スポット以外のスポ

It is selective for H1 receptors over H2 and H3 receptors (Kis = 987 and 2,500 nM, respectively). Ketotifen (50 and 100 μM) inhibits degranulation of rat peritoneal mast cells induced by compound 48/80 . It inhibits the passive cutaneous anaphylaxis (PCA) reaction in rats by 54.6% when administered orally at a dose of 20 mg/kg. Ketotifen (30 mg/kg) inhibits the quick phase airway response in a rat ovalbumin-induced immediate airway response model. The solution contains0.345 mg of ketotifen fumarate, which is equivalent to0.25 mg of ketotifen. The solution also contains the preservativebenzalkonium chloride (0.01%) as well as glycerol,sodium hydroxide and/or hydrochloric acid (to adjust pH),and purified water. It has a pH of 4.4 to 5.8 and an osmolalityof 210 to 300 mOsm/kg.
The recommended dose of ketotifen solution is 1 drop instilledinto each affected eye every 8 to 12 hours. The mostfrequently reported adverse reactions are conjunctival infection,headaches, and rhinitis.
