Zalcitabine (2′-3′-dideoxycytidine, ddC), also called dideoxycytidine, is a nucleoside analog reverse transcriptase inhibitor (NARTI) sold under the trade name Hivid. Zalcitabine was the third antiretroviral to be approved by the Food and Drug Administration (FDA) for the treatment of HIV/AIDS. It is used as part of a combination regimen.

Zalcitabine appears less potent than some other nucleoside RTIs, has an inconvenient three-times daily frequency and is associated with serious adverse events. For these reasons it is now rarely used to treat human immunodeficiency virus (HIV), and it has even been removed from pharmacies entirely in some countries.

Production method

N-acetyl cytosine nucleoside (4g, 14mmo1) was refluxed in 200ml acetonitrile, acetyl bromide (10ml, 0.137mo1) was added within 30min, and then refluxed for 3h. Steam out the solvent, the remaining soluble in 100ml chloroform, 100ml water. The remaining material was recrystallized with 10ml ethanol to obtain 2g colorless flake crystalline compound (I) with 33% yield and melting point of 179-180℃(decomposition). Compound (I)(1g, 2.3mmo1), lg calcium carbonate, 0.2g palladium-barium sulfate and 50ml 50% methanol were shaken with hydrogen gas at 25℃ and atmospheric pressure for 1h(to absorb 60ml hydrogen). Filter to remove the crystals and concentrate the filtrate until dry. The residue was dissolved in 10ml 20% ammonia-methanol solution and placed at 5℃ overnight. After column chromatography and recrystallization with ethanol, 34mg colorless crystalline Zaxitabine was obtained with yield of 7%.
