Reaction Conditions:
Stage #1: 3-ethoxysalicylaldehydewith sodium chlorite;sodium dihydrogen phosphate monohydrate;aminosulfonic acid in 1,4-dioxane;water at 0 - 20; for 2 h;
Stage #2: with sodium sulfite in 1,4-dioxane;water at 20; for 0.5 h;
Steps:
1.1 Step 1:
To a solution of 3-ethoxy-2-hydroxybenzaldehyde (5g, 30.120mmol, 1 eq) in 1 ,4-dioxane:H20 (4:2) (60ml) was added sulfamic acid (4.4g, 45.180, 1.5eq), NaH2P04.H20 (16.2g, 1 17.46mmol, 3.9eq), and NaCI02 (3.52g, 39.15mmol, 1.3eq) at 0°C.The reaction mixture was allowed to warm up to RT then stirred for 2h at rt. Then Na2S03 (4.55g, 36.144mmol, 1.2eq) was added at RT and stirred for 30 min at RT. TLC analysis indicated formation of a polar spot. The reaction mixture was acidified with Con.HCI (10ml_) and extracted with EtOAc (50ml_ X 3). The combined organic layer was dried over Na2S04 and concentrated under reduced pressure to give a crude product. The crude product was purified by solvent wash to afforded analytically pure 3- ethoxy-2-hydroxybenzoic acid (5.3g, 96.7%) as an off white solid. LC-MS: m/z 181.19 (M- H).
