Reaction Conditions:
Stage #1:acetyl chloride with aluminum (III) chloride in 1,2-dichloro-ethane at 0 - 10; for 1 h;
Stage #2:2-oxoindole in 1,2-dichloro-ethane at 0 - 20;
Steps:
1.1; 2.1; 3.1 (1) Synthesis of 5-acetylindole-2-one (2):
Add 30.0g AlCl3 and 100mL dichloroethane to a 250mL three-necked flask. After the ice bath is cooled to 0, 11.8g of acetyl chloride is added dropwise and the temperature is controlled not to exceed 10. After the addition is completed, keep stirring for 1h, and then add 10g 2- Indolinone and control the temperature at 0-15 , after the addition, slowly return to room temperature and stir overnight. After the reaction is completed, slowly add the reaction solution to 400g of ice water under vigorous stirring and stir for 30 minutes. The filtered solid is washed with water and dried. 12.4 g of 5-acetylindol-2-one was obtained with a yield of 94.3%.
