41.1 Step 1: 7-methylciuinazoline-2.4( 1 H.3H)-dione

A mixture of 4-methyl-2-aminobenzoic acid (2.00 g, 13.23 mmol) and urea (8.00 g, 133.21 mmol) was heated to 160°C and stirred for 10 h, then cooled to 100°C. To the reaction mixture was added water (100 mL), and the resulting mixture was stirred for 5 mm. After the reaction, the mixture was filtered and the filter cake was washed with water (100 mL). The filter cake was added into a sodium hydroxide solution (0.5 M, 100 mL), and the mixture was kept boiling for 10 mm. Then the mixture was cooled, and acidified with diluted hydrochloric acid (1 M) to pH about 1 to 2. The resulting mixture was filtered, and the filter cake was washed with water (100 mL) to give the title compound as a yellow solid (2.30 g, 99%).
