Reaction Conditions:
Stage #1: 2-(1,3-dioxo-1,3-dihydro-isoindol-2-yl)ethanesulfonic acid dimethylamidewith hydrazine hydrate in ethanol; for 7 h;Reflux;
Stage #2: with hydrogenchloride in water;
Steps:
General procedure: To a solution of M-3 (0.80 g, 4.00 mmol) in ethanol (30 mL), hydrazine hydrate (80%, 0.30 g, 4.80 mmol) was added. After refluxed for 7 h, a white precipitate formed that was removed by filtration. The filtrate was evaporated to dryness and added to water (100 mL). The aqueous suspension was acidified with conc. HCl, and residual insoluble material was filtered off. The clear filtrate was evaporated to give amine-17. Amine-19 was synthesized as the same procedure of amine-17. Yield: 90.3%; 1H NMR (400 MHz, D2O) δ 3.45-3.40 (m, 4H), 2.82(s, 6H).
