5 4.2.5 1-(4-(benzylamino)-7,8-dihydro-5H-pyrano[4,3-d]pyrimidin-2-yl)-2-methyl-1H-indole-4-carboxamide (CB-5083)
General procedure: At room temperature, to the solution of the 16 (262mg, 0.663mmol) in ethanol (12mL) was added PPh3 (34mg, 0.13mmol), Pd(OAc)2 (29.2mg, 0.13mmol), acetaldehyde oxime (35.4mg, 0.60mmol) and water (1.5mL). The resulting mixture was refluxed for 2h, cooled down to room temperature and concentrated in vacuo. The residue was purified by flash chromatography (petroleum ether/EtOAc=2:1) to afford CB-5083 (191mg, yield 70%, purity 99%).
