Steps:

l-Xylulose production using recombinant HjLAD
The initial reaction mixture contained 5 g·L-1 of l-arabinitol and 20 mM of NAD+. To start the reaction, 20 U·mL-1 of purified HjLAD was added. The reaction was performed at 25°C and pH 9.5 or 7.0 in a 3-mL thermoreactor with a stir bar. Samples were withdrawn at constant time intervals and analyzed by the cysteine carbazole sulfuric method and the absorbance was measured at 560 nm.4 The results were further confirmed by HPLC, an Ultimate 3000 high-pressure liquid chromatography system (Dionex, Sunnyvale, CA, USA) equipped with a Shodex sugar sp0810 column (Showa Denko, K. K., Kawasaki, Japan) and an evaporation light scattering detector (ESA6700, Chromachem, MA, USA). Samples were eluted with water at a rate of 1 mL·min-1 at 80°C (column temperature). The nebulizer and evaporation temperatures were 50°C and 65°C, respectively. The retention times for l-arabinitol and l-xylulose were 16.1 and 9.8 min, respectively, at operating conditions (data not shown).
