2.6.2 Immobilized enzyme
Immobilized enzyme activity was measured by incubating 0.025 g of immobilized enzyme with 1mL of 40 g/L (w/v) sucrose in buffer B at 30 °C for 20 min and stopped by centrifuging at 12,000×g for 1 min. Thus, 1U of immobilized enzyme activity is defined as the amount of SIase that releases 1μmol isomaltulose per minute under standard assay conditions.
