Steps:

Synthesis of N-phthaloyl-DL-glutamic acid (8)

A mixture of phthalic anhydride 3 (112 g, 0.75mol), L-glutamic (110 g, 0.75mol) and pyridine(75mL) in a round-bottom flask was subjected to microwave irradiation (100 W,115 C, 15min) with stirring. After the reaction was finished, the round-bottom flask wasremoved from the microwave apparatus. The reaction mass was cooled to 75 C, ice-coldwater (900mL) was added, the pH was adjusted to 1.2 with 6N HCl and the mixture wasstirred at 10-15 C over 2 h. The resulting white solid was separated, filtered, and washedwith cold water (3 x 40mL). The obtained product was dried under vacuum to afford Nphthaloyl-DL-glutamic acid (8) (186.7 g, 89.34%), mp: 191-193 C Rf 0.41 (benzene:dioxane:formic acid 75:20:5), with the expected spectrometric features (see SupplementaryMaterials). This material was sufficiently pure to be used directly in the next step.
