(R,S)-Amlodipine (100.00g, 244.6mmol) was dissolved in a mixed solvent of N,N-dimethylformamide and purified water (VDMF: Vwater= 7:1, 900 mL), and filtered Insoluble impurities were removed. After the filtrate was stirred at room temperature for 30 minutes, L-tartaric acid (18.40g, 122.6mmol) of N,N-dimethylformamide and purified water (VDMF:Vwater= 7:1, 300mL) ), after the dropwise addition is completed, stir at room temperature for 50 min.Add a small amount of S-(-)-amlodipine-L-tartrate seed crystals and continue to stir for 4h to crystallize.The white solid was filtered, washed with cold acetone (150 mL), and dried under pressure and vacuum for 10 hours to obtain 50 g of S-(-)-amlodipine-L-tartrate. The molar yield was 42.24percent; the HPLC optical purity was 99.49percent.The S-(-)-Amlodipine-L-tartrate (50g, 103.3mmol) obtained above was directly dissolved in a mixed solution of water and isopropanol (Vwater: Visopropanol=4:1, 300mL) Add benzene sulfonic acid aqueous solution (16.5g dissolved in 45mL purified water), stir to crystallize, filter, the filter cake is washed with water (200mL×2), then blow dried at 40, it will be white or almost white45.1g oflevoamlodipine besylate solid, with a molar yield of 77.0percent. According to HPLC purity test, the one with tR=12.023min is levamlodipine besylate with a purity of 99.44percent, tR=7.750min Impurity D is 0.49percent; the optical purity detected by HPLC is 99.55percent.
