Hazard Information | Back Directory | [Synthesis]
GENERAL METHOD: To a mixture containing 2-phenylimidazo[1,2-a]pyridine (1a, 39 mg, 0.2 mmol), molecular iodine (I2, 50 mg, 0.2 mmol), and 4-(3-methylimidazolyl)butane-1-sulfonate (I, 9 mg, 0.04 mmol), water (2 mL) was added. The mixture was transferred to a sealed tube and stirred at room temperature for 2 hours. After completion of the reaction (monitored by TLC), the mixture was extracted with ethyl acetate (20 mL). The organic phase was dried with anhydrous sodium sulfate (Na2SO4) and concentrated under reduced pressure to give the crude product. The crude product was purified by column chromatography (silica gel, 60-120 mesh, petroleum ether/ethyl acetate, 9:1) to give 2-iodo-1-methyl-1H-imidazole (2a) as a white solid. Yield: 58 mg (91%); melting point: 132-134 °C. | [References]
