向配备有磁力搅拌子的1L圆底烧瓶中加入187.8g(1mol)5-羟基间苯二甲酸(纯度约97%,可购自Aldrich)和500mL(12.3mol)甲醇。缓慢加入28mL浓硫酸后,将反应混合物加热至回流温度并在回流条件下搅拌4小时。反应完成后,将热反应液小心倒入500mL冰水中。随后,通过过滤收集白色固体产物,并用去离子水多次洗涤直至洗涤液呈中性。将产物在60℃下真空干燥过夜,得到206.8g 5-羟基间苯二甲酸二甲酯。产物表征数据如下:熔点164-166℃;GC纯度>99%;收率98.2%;1H NMR(CDCl3 + DMSO-d6,TMS为内标,ppm):3.68(s,6H,OCH3),7.45(s,2H,Ar-H),7.89(s,1H,Ar-H),9.35(宽峰,1H,OH);13C NMR(CDCl3 + DMSO-d6,TMS为内标,ppm):51.92(OCH3),120.50(C4,C6),121.10(C2),131.15(C1,C3),157.36(C5),165.85(COOR);1H-13C NMR相关谱(CDCl3 + DMSO-d6,TMS为内标,ppm):51.92(四重峰,OCH3,1JCH = 147Hz),120.50(双峰,C4,C6,1JCH = 164.4Hz),121.10(双峰,C2,1JCH = 168.3Hz),131.15(单峰,C1,C3),157.36(单峰,C5),165.85(单峰,COOR);GC-MS(丙酮为溶剂):m/z 210(M+)。
