一般步骤:将N-α-芴甲氧羰基-N-ε-叔丁氧羰基-L-赖氨酸(5.66 g,12.1 mmol)溶于4 M HCl/二恶烷(120 mL)中,室温搅拌2小时以脱除叔丁氧羰基保护基。反应完成后,减压浓缩除去溶剂。将所得残余物溶于乙醇(60 mL)中,随后加入2-(1-羟基亚乙基)-5,5-二甲基环己烷-1,3-二酮(3.36 g,18.4 mmol)和N,N-二异丙基乙胺(6.2 mL,35.6 mmol)。将反应混合物加热回流17小时。冷却至室温后,减压浓缩除去溶剂。将残余物溶于乙酸乙酯(300 mL)中,依次用1 M HCl(100 mL)和饱和食盐水(100 mL)洗涤,有机相用无水硫酸钠干燥,过滤并浓缩。通过硅胶柱色谱法(0.5%-3%甲醇/二氯甲烷梯度洗脱)纯化粗产物,得到N-芴甲氧羰基-N'-[1-(4,4-二甲基-2,6-二氧代环己亚基)乙基]-L-赖氨酸(2.64 g,收率41%),为白色固体。产物光谱数据与文献报道一致。1H NMR (500 MHz, CDCl3): δ 13.31 (br s, 1H), 7.75 (d, J = 7.8 Hz, 2H), 7.59 (t, J = 7.8 Hz, 2H), 7.38 (t, J = 7.8 Hz, 2H), 7.31-7.28 (m, 2H), 5.73 (d, J = 8.0 Hz, 1H), 4.48-4.45 (m, 1H), 4.37 (d, J = 7.1 Hz, 2H), 4.20 (t, J = 7.1 Hz, 1H), 3.43-3.40 (m, 2H), 2.55 (s, 3H), 2.36 (s, 4H), 2.00-1.50 (m, 6H), 1.01 (s, 6H). HR-MS (m/z, FAB): 计算值C31H37N2O6 ([M+H]+), 533.2652; 实测值, 533.2643.
